Fluorescence Imaging Study of Film Coating Structure and Composition Effects on DNA Hybridization

Hybridization of surface-bound DNA with complementary strands is the basis many biotechnological applications. Herein, structure interfacial coatings between substrate and bound a crucial element for hybridization behavior. three reactive surfaces constructing DNA-sensing platforms, namely, plain gold films on silicon, poly(bisphenolA-co-epichlorohydrin) (PBAG) brush-like bilayer structure, dibenzocyclooctyne monolayers (both glass), are compared. Fluorescence imaging employed to survey effect coating conformation performance. To better understand structural properties chemistry coated films, atomic force microscopy, water contact angle measurements, X-ray photoelectron spectroscopy characterize surface morphology. probe microarrays created different platforms via microchannel cantilever spotting, their performance hybridizing counterparts assessed. While all work reliable detection, protein-binding assay reveals that PBAG offer highest efficiency among these approaches. The results present have significant implications comprehension interactions physico-chemical can inform fabrication sensors.
